All Chrom Z kits can be used in endpoint determinations on any spectrophotometer at 405 nm. Recommended is the PACKS-4 semi-automated, kinetic method which allows use of smaller sample and reagent volumes.

The Chrom Z Factor VIII:C assay is useful in diagnosing Hemophilia A, monitoring therapy, and QC of Factor VIII concentrate and cryoprecipitate products. After activation by thrombin, F.VIII:C acts as a cofactor in conversion of F.X to F.Xa in the presence of F.IXa, calcium and phospholipids. F.Xa generated is measured using a specific chromogenic substrate and is directly proportional to F.VIII:C in the sample.

Chrom Z Heparin is for quantitative determination of therapeutic heparin (standard and low molecular weight) in human plasma. In presence of F.Xa and excess AT III, the rate of F.Xa inhibition is directly proportional to heparin concentration. Residual F.Xa activity, measured with a F.Xa-specific chromogenic substrate, is inversely proportional to heparin concentration.

Chrom Z Plasminogen is useful in detecting congenital and acquired deficiencies, particularly DIC, and in monitoring thrombolytic therapy. Specificity of the Chrom Z method is ensured by use of a chromogenic substrate specific for the streptokinase-plasminogen complex or plasmin, and by use of streptokinase which has no proteolytic activity by itself and is known to activate only plasminogen in human plasma.

This Chrom Z assay is for quantitative determination of Protein C (PC). When activated, PC inhibits coagulation by inactivating F.Va and F.VIIIa, and has also been shown to stimulate fibrinolysis. PC in plasma is rapidly and selectively activated by using snake venom. Activated PC is detected by monitoring hydrolysis (release of pNA) of a PC-specific chromogenic substrate.
